Although surrogate assays have been developed to investigate SARS-CoV-2 variant specific features, truly virological studies definitely need live virus. For example, analyzing cell and tissue tropism and replication kinetics, antiviral drug discovery and testing, and measuring antibody response are all applications best run with live virus.
